地球を含む複数の惑星で構成される惑星連邦が設立されて約1世紀。西暦2264年、未知の惑星の調査や知的生命体との友好的接触を目的として、惑星連邦の宇宙艦隊が誇る全長２８９メートル、コンスティテューション級の宇宙船U.S.S.エンタープライズNCC-1701が４３０名の乗組員を乗せ、人類未踏の地－宇宙へ旅立った。エンタープライズ号を率いるのは、３０歳の若さで船長に抜擢された不屈の精神力を持つ男ジェイムズ・T・カーク。地球人と異星人バルカン人の間に生まれ理性を重んじる副長のスポックや、人間味溢れる医療主任のドクター・マッコイといった優秀なクルーの力も得て、エンタープライズ号は広大な宇宙空間で起こる想像もできないような神秘現象、高度な文明を持つ生命体との遭遇などさまざまな冒険を繰り広げるのだ。
