カーク船長やミスター・スポックたちエンタープライズ号のクルーによる、伝説の調査航海から約1世紀後の24世紀。地球を中心とする150の文明圏で構成される惑星間同盟“惑星連邦”は宇宙艦隊を組織、未知宇宙の探査と連邦の防衛・パトロールに当たらせていた。新造艦エンタープライズ号は以前より大きく、全長は2倍以上に、内部スペースは8倍にもなっている。乗組員には、クルーの家族、子供も含まれており、総勢2000人を数える。
これらの乗組員や新しくなったエンタープライズ号を率いるのは宇宙艦隊のジャン＝リュック・ピカード大佐。彼は常に冷静であり、困難に直面したときほど優れたリーダーシップを発揮する。そして、彼の下で指揮をとる副長のライカー中佐は、オリジナル・シリーズのカーク船長の性格を受け継ぐ熱血漢ぶりを発揮する。また生物の感情を関知する能力を持つベタゾイド人のディアナ・トロイ、クリンゴン人のウォーフ、人間を超える能力を持ちながら人間に憧れる艦隊で唯一のアンドロイドのデータ少佐など、個性的なクルーたちが素晴らしいチームワークを発揮し、未知なるものを求めて広大な宇宙の旅を続けていくのだ。
